Changes in Toyal Circulating Blood Volume and Red Cell Volume of the Various Organs in Rats Induced by Ethanol Administration

Abstract
The rats were orally administered a single dose of lml (approximately equivalent to 2gm of pure ethanol per kg body weight) or 3m1. of 33% ethanol. The effects of ethanol on the total circulating blood volume, red cell volume of spleen, kidney, liver and lung were measured.
In this study 77 rats were divided into control and experimental groups, and the later was further divided into lml. group and 3m1. group according to the dose of oral administration.
Observations were made at lOmin., 30min., 1 hour, 3 hours and 5hours after ethanol administration.
Total circulating blood volume was measured using the dilution principle with 61Cr labeled red cell, and from body hematocrit, red cell volume and plasma volume were derived.
Red cell volume of each organ of the animal was calculated from c. p. m. of ~¢¥¢¥Cr in the lml. of a sample blood of the animal.
The results obtained were sur,2marized as follows
1. In the lml. group, total circulating blood volume decreased as the time progressed after the administration of ethanol up to 1 hour, buta gradual recovery set in thereafter. In the 3m1. group, after a tendency of minor increase during the first hour, the volume decreased until the third hour, and sudsequetly recovered.
2. In all the experiments, red cell volume of each organ increased at 30min. after ethanol administIn the 3m1. group, red cell volume increased somewhat: at a later hour in the liver.
However, in both groups, the red cell volume of the spleen increased during the initial 30 min. and then markedly decreased thereafter.
3. The reason for the difference of the early and later organ red cell volume changes between the lml. group and the 3 ml. group except in the spleen, is presumed to be caused by the influence of a central cardiovascular depressant effect.
